4.4. Isolation and identification of Rhizoctonia fungi on PDA medium
( Rhizoctonia re-isolation)
The evaluation of the colonization of the stem bases was supplemented by a mycological analysis, which was performed after collecting the emitted volatile compounds. The material for the mycological analyses was taken from the wheat stem base with disease symptoms (if visible), regardless of the experimental treatment. From each wheat plant, one 10 mm section from the diseased stem bases was prepared. The stem pieces were rinsed for 30 min in tap water, disinfected for 20 s in 1% AgNO3 solution and then rinsed three times in sterile distilled water and placed onto potato dextrose agar (PDA, Difco +) in Petri dishes with 50 mg of streptomycin per 1 dm3 and incubated for 14 days at 22 ◦C. Cultures of fungi were identified by their morphology on PDA and synthetic nutrient agar (SNA) (Lemanczyk´and Kwa´sna, 2013).
